Bu çalışmada, Damascus (DMS), Kilis (KLS), Hatay (HTY) ve Kıl (KIL) keçi
ırklarında mitokondriyal DNA çeşitliliğinin D-loop üzerinde 598 bç uzunluğundaki bölgenin
DNA dizi analizi ile ortaya konması amaçlanmıştır. Çalışmanın hayvan materyalini T.C.
Tarım ve Orman Bakanlığı, Tarımsal Araştırmalar ve Politikalar Genel Müdürlüğü tarafından
yürütülen “Halk Elinde Hayvan Islahı” ülkesel projesi kapsamında yetiştirilen keçiler
oluşturmuştur. Bu amaçla toplamda 84 baş keçiden kan örnekleri alınmış ve DNA dizi analizi
ile değerlendirmeler yapılmıştır.
Dört farklı ırk ve toplamda 84 baş keçide 65 haplotip, 95 polimorfik bölge ve 64
parsinomik bölge tespit edilmiştir. Haplotip çeşitliliği en yüksek Damascus (H=0,9890,019)
ve en düşük ise Hatay (H=0,9440,028) keçilerinde hesaplanmıştır. Nükleotid çeşitliliği
bakımından en yüksek ve düşük değerler sırasıyla Damascus (=0,026350,00336) ve Kıl
(=0,017340,00109) keçi ırklarında olduğu gözlenmiştir. Popülasyon genelinde 80 baş
hayvanın A haplogrubunda, dört baş hayvanın ise G haplogrubunda olduğu belirlenmiştir.
Daha önce literatürde yer almayan Hatay keçilerinin A ve G haplogruplarında olduğu tespit
edilmiş, aynı zamanda Damascus keçilerinde ise G haplogrubu ilk kez bu çalışmada
belirlenmiştir. Nötralite test sonuçlarına göre özellikle Hatay keçi popülasyonunda Fu ve
Li'nin D ile F testi sonuçlarının, nadir allellerin azlığına ve popülasyonda dar boğazın hafif
etkilerine işaret ettiği görülmektedir. Nei’nin genetik uzaklık matrisine göre çizdirilen
dendogram, Kilis keçilerinin bölgedeki Kıl keçi ve Damascus keçileri ile kontrolsüz şekilde

melezlendiğini, bununla birlikte Hatay keçilerinin ise Kilis ve Kıl keçisi ile melezlenerek
ortaya çıktığı bilgisini desteklemektedir.
Sonuç olarak, üzerinde çalışılan ırklar bakımından mitokondriyal DNA D-loop
bölgesinin ve yapılan analizlerin genetik varyasyonu ortaya koyması açısından yeterli olduğu
söylenebilir. Elde edilen bulguların literatüre önemli katkılar sağlayacağı düşünülmektedir. |

This study aims to investigate the mitochondrial DNA diversity in Damascus (DMS),
Kilis (KLS), Hatay (HTY), and Kıl (KIL) goat breeds through DNA sequence analysis of the
598 bp D-loop region. The animal material for this study comprised goats raised under the
national project “On-Farm Animal Breeding,” conducted by the General Directorate of
Agricultural Research and Policies of the Ministry of Agriculture and Forestry of the
Republic of Türkiye. For this purpose, blood samples were collected from a total of 84 goats,
and evaluations were carried out through DNA sequence analysis.
In four distinct breeds and a total of 84 goats, 65 haplotypes, 95 polymorphic regions,
and 64 parsimony-informative regions were identified. The highest haplotype diversity was
observed in the Damascus breed (H=0,989±0,019), while the lowest haplotype diversity was
recorded in the Hatay goats (H=0,944±0,028). In terms of nucleotide diversity, the highest
and lowest values were observed in the Damascus (π=0,02635±0,00336) and Kıl
(π=0,01734±0,00109) goat breeds, respectively. Across the population, 80 animals were
found to belong to haplogroup A, while four animals belonged to haplogroup G. It was
determined that the Hatay goats, which were not previously documented in the literature,
belonged to both haplogroups A and G. Additionally, haplogroup G was identified in the
Damascus goats for the first time in this study. According to the neutrality test results,
especially in the Hatay goat population, the outcomes of Fu and Li's D and F tests indicate a
deficiency of rare alleles and imply a slight bottleneck effect within the population.

According to the Phylogram tree constructed based on Nei's genetic distance matrix,
the results support the idea that Kilis goats have been uncontrollably mated with local Kıl
and Damascus goats. Additionally, the data support the idea that Hatay goats have emerged
through mating with Kilis and Kıl goats.
In conclusion, the mitochondrial DNA D-loop region and the analyses conducted were
sufficient to reveal the genetic variation among the studied breeds. The findings are expected
to make significant contributions to the existing literature. |
