Amaç: Bu çalışma, Aydın ilinin çeşitli ilçelerindeki farklı mandıralarda üretilip satışa sunulan tulum ve beyaz peynirlerde Staphylococcus aureus (S. aureus) varlığının araştırılması ve izole edilen izolatlardan klasik stafilokokal enterotoksin genlerinin (sea - see) belirlenmesi amacıyla gerçekleştirilmiştir. Gereç ve Yöntem: Bu çalışmada 2021 yılında Aydın ilinin çeşitli ilçelerindeki (Çine, Efeler, İncirliova, Köşk, Kuşadası, Nazilli, Söke) farklı mandıralarda üretilen ve satışa sunulan 100 adet peynir örneği (50 tulum peyniri, 50 beyaz peynir) steril poşetler içerisinde, soğuk zincir altında Aydın Adnan Menderes Üniversitesi Veteriner Fakültesi Besin Hijyeni ve Teknolojisi Anabilim Dalı Laboratuvarına getirilerek S. aureus ve klasik enterotoksin genlerinin varlığı yönünden incelemeye tabi tutulmuştur. Bulgular: Çalışma kapsamında incelenen 100 adet peynir örneğinin 18 tanesinin S. aureus ile kontamine olduğu tespit edilmiştir ve 18 örneğin 18’inin (%100) PCR ile doğrulanması gerçekleştirilmiştir. Analizleri yapılan 50 adet tulum peyniri örneklerinden 9’unun (%18) ortalama 4,92 log kob/g düzeyinde, 50 adet beyaz peynirin 9’unun (%18) ortalama 5,56 log kob/g düzeyinde, tüm peynir örneklerinin ortalama 5,24 log kob/g düzeyinde S. aureus ile kontaminasyonu tespit edilmiştir. Tulum peyniri örneklerine ait S. aureus izolatlarından 4 tanesinin (%44,4), beyaz peynir örneklerinden 2 tanesinin (%22,2), toplamda ise 6 tanesinin (%33,3) klasik enterotoksin genlerinden (sea, seb, sec, sed, see) en az birine sahip olduğu belirlenmiştir. Toplamda izole edilen 6 enterotoksijenik izolatta 3 tane sea, 3 tane sed ve 2 tane seb geni bulunmuştur. Peynir izolatlarının hiçbirinde sec ve see genlerinin varlığı saptanmamıştır. Sonuç: İncelenen peynir örneklerinin S. aureus ile çeşitli düzeylerde kontamine olduğu tespit edilmiştir ve S. aureus içeren peynir izolatlarının bir kısmının klasik enterotoksin genlerini taşıdığı belirlenmiştir. İncelenen peynirlerde gıda hijyeninin yeterli düzeyde olmadığı, örneklerde belirlenen enterotoksinlerin gıda zehirlenmelerine sebebiyet verebileceği ve halk sağlığı açısından tehdit oluşabileceği sonucuna ulaşılmıştır.
Objective: This study was carried out to investigate the presence of Staphylococcus aureus (S.
aureus) in tulum and white cheese produced and sold in different dairy farms in various districts
of Aydın and to determine the classical staphylococcal enterotoxin genes (sea - see) in isolated
strains.
Material and Methods: In this study, 100 cheese samples (50 tulum cheese, 50 white cheese)
produced and offered for sale in different dairy farms in various districts of Aydın (Çine, Efeler,
İncirliova, Köşk, Kuşadası, Nazilli, Söke) in 2021 were packed in sterile bags, and brought to
Aydın Adnan Menderes University Faculty of Veterinary Medicine Department of Food
Hygiene and Technology Laboratory under cold chain and examined for the presence of S.
aureus and classical enterotoxin genes.
Results: Eighteen of 100 cheese samples examined within the scope of the study were found
to be contaminated with S. aureus, and 18 of 18 samples (100%) were confirmed by PCR. 9 of
50 tulum cheese samples (18%) were analyzed at an average of 4.92 log cfu/g, 9 of 50 white
cheeses (18%) were at an average of 5.56 log cfu/g, and contamination of all cheese samples
with S. aureus was determined at an average level of 5.24 log cfu/g. It was determined that 4
of the S. aureus isolates of tulum cheese samples (44.4%), 2 of the white cheese samples
(22.2%), and 6 of them in total (33.3%) had at least one of the classical enterotoxin genes (sea,
seb, sec, sed, see). In total, it was determined that 6 enterotoxigenic isolates carried 3 of the sea,
3 of the sed, and 2 of the seb genes. The presence of sec and see genes was not detected in any
of the cheese isolates.
Conclusion: It was determined that the cheese samples examined were contaminated with S.
aureus at various levels and some of the cheese isolates containing S. aureus were found to

carry classical enterotoxin genes. It has been concluded that the food hygiene in the cheese
samples examined is not at a sufficient level, that the enterotoxins determined in the samples
may cause food poisoning and may pose a threat to public health.
