Amaç: Gerek terapötik açıdan gerekse önemli bir protein kaynağı olmasından dolayı besleyici öneme sahip deve sütünün inek sütüne göre serum proteini ve kazein oranlarının ile protein profilinin farklı olması bu sütün fermente süt ürünlerine işlenebilmesinde zorluklar yaratmaktadır. Bu nedenle bu çalışmada deve sütünün inek sütüyle karıştırılarak kefir üretiminde kullanımı hedeflenmiştir. Ayrıca çalışmada istenilen viskozitede kefir üretimini sağlamak amacıyla mikrobiyal transglutaminaz enzimi kullanımının etkisi araştırılmıştır. Materyal ve Yöntem: Çalışmada çiğ deve ve inek sütlerine fiziksel ve kimyasal analizler uygulanarak deve ve inek sütlerinin %80 : %20 ve %70 : %30 olduğu karışımlar kefir üretimi için seçilmiş olup yanıt yüzey metoduyla kefir starter kültür oranı ile mTGaz konsantrasyonu varyasyona tabi tutulmuştur. Denemeler sonucunda en yüksek viskoziteye sahip starter kültür ve enzim konsantrasyonlarından 4 farklı kefir üretimi yapılmış ve mTGaz ilave edilmeden üretilmiş 2 kefir örneğiyle sodyum dodesil sülfat-poliakrilamid jel elektroforezi, depolama analizleri, mikroyapı, uçucu bileşenler ve duyusal açıdan karşılaştırılmıştır. Bulgular: Üretim için seçtiğimiz 6 adet kefir örneğinin 24 saatlik inkübasyon sonunda relatif vizkoziteleri; %70 deve sütü %30 inek sütü %0,2 starter kültür 3 U/g protein mTGaz ilaveli örnekte 33,60; %70 deve sütü %30 inek sütü %0,17 starter kültür 5,12 U/g protein mTGaz ilaveli örnekte 66,21; %70 deve sütü %30 inek sütü %0,1 starter kültür 0 U/g protein mTGaz ilaveli örnekte 1,85; %80 deve sütü %20 inek sütü %0,2 starter kültür 3 U/g protein mTGaz ilaveli örnekte 31,25; %80 deve sütü %20 inek sütü %0,17 starter kültür 5,12 U/g protein mTGaz ilaveli örnekte 26,30; %80 deve sütü %20 inek sütü %0,1 starter kültür 0 U/g protein mTGaz ilaveli örnekte ise 2,68 olmuştur. mTGaz ilavesiyle üretilen kefir örneklerinde sodyum dodesil sülfat-poliakrilamid jel elektroforezi analizi sonucunda, mTGaz konsantrasyonunu artışıyla proteinlerin polimerizasyon oranının artışı kazein bantlarının yoğunluğunun azalması üzerinden yorumlanmıştır. Depolama analizleri sonucunda su tutma kapasitesi ve serum ayrılmasının depolama süresince arttığı görülmüştür. Serum ayrılması ve su tutma kapasitesinin depolama süresine bağlı değişimi istatiksel açıdan anlamlı bulunmuştur (P> 0,05). Kullanılan süt karışımı, starter kültür oranı ve mTGaz konsantrasyonuna bağlı olarak kefirlerde oluşan uçucu bileşen miktarlarının ve çeşitlerinin değiştiği saptanmıştır. mTGaz ilave edilen kefirlerin mikro yapısının mTGaz ilave edilmeyen kefirlere göre daha sıkı olduğu tespit edilmiştir. Üretilen kefir örneklerinin tamamı %laktik asit (en az %0,6) oranı bakımından Türk Gıda Kodeksi Fermente Süt Ürünleri Tebliği (Tebliğ Numarası 2009/25)' ne uygun bulunmuştur. Kefir örneklerine yapılan duyusal analiz sonucunda örneklerin tamamı panelistlerce beğenilmiştir. Sonuç: Deve sütünün inek sütüyle karıştırılarak mTGaz ilavesiyle kefir üretiminin yapılması kefirlerin bazı özelliklerini iyileştirmiş ve mTGaz ilavesinin deve sütünden kefir üretiminde umut vadedici iyileştirme sağladığını göstermiştir.
Objective: The fact that the composition of camel milk, which has nutritional importance due to its therapeutic and important protein source, is different from cow's milk, creates difficulties in processing this milk into fermented milk products. Therefore, in this study, it is aimed to use camel milk mixed with cow's milk in kefir production. In addition, the effect of using microbial transglutaminase in order to produce kefir with the desired viscosity was investigated in the study. Material and Method: In the study, physical and chemical analyzes were applied to raw camel and cow's milk, and mixtures of 80%: 20% and 70%: 30% of camel and cow's milk were selected for kefir production, and kefir starter culture ratio and mTGase concentration were varied by response surface method. As a result of the experiments, 4 different kefirs were produced from the starter culture and enzyme concentrations with the highest viscosity, and 2 kefir samples produced without adding mTGase were compared with sodium dodecyl sulfate- polyacrylamide gel electrophoresis, storage analysis, microstructure, volatile components and sensorial properties. Results: Relative viscosities of 6 kefir samples selected for production after 24 hours of incubation; 70% camel milk 30% cow milk 0.2% starter culture 3 U/g protein 33.60 in the sample with mTGase added; 70% camel milk 30% cow milk 0.17% starter culture 5.12 U/g protein 66.21 in the sample with mTGase; 70% camel milk 30% cow milk 0.1% starter culture 0 U/g protein 1.85 in the sample with mTGase added; 80% camel milk 20% cow milk 0.2% starter culture 3 U/g protein 31.25 in the sample with mTGase added; 80% camel milk 20% cow milk 0.17% starter culture 5.12 U/g protein 26.30 in the sample with added mTGase; 80% camel milk 20% cow milk 0.1% starter culture 0 U/g protein in the sample with mTGase added, it was 2.68. As a result of sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis in kefir samples produced with the addition of mTGase, it was observed that the polymerization rate of proteins increased with the increase in mTGase concentration, and the density of casein bands decreased. As a result of storage analysis, it was observed that water holding capacity and serum separation increased during storage. The changes in serum separation and water holding capacity depending on the storage time were found to be statistically significant (P> 0.05). It has been observed that the amount and types of volatile components formed in kefir change depending on the milk mixture used, starter culture ratio and mTGase concentration. It has been determined that the microstructure of kefirs with added mTGase is tighter than kefirs without enzyme. All of the kefir samples produced were found to comply with the Turkish Food Codex Communiqué on Fermented Dairy Products (Communiqué Number 2009/25) in terms of %lactic acid (at least 0.6%). As a result of the sensory analysis performed on kefir samples, all of the samples were appreciated by the panelists. Conclusion: The production of kefir by mixing camel milk with cow's milk and adding mTGase improved some properties of kefir and demonstrated that mTGase is a promising technique in the production of kefir from camel milk.
