Glioblastoma multiform, glia hücrelerinden türeyen primer beyin tümörüdür. Tümörün
büyümesi, glioma hücrelerini çevreleyen nöronların eksitotoksisite yolu ile ölmesiyle
gerçekleşir. Eksitotoksisite, sinaptik boşlukta aşırı glutamat birikimi ile oluşur ve epilepsi,
felç ve nörodejeneratif hastalıklar gibi beyin hastalıkları ile glioblastoma gibi beyin
tümörlerinde görülür. Glutamat, merkezi sinir sisteminde birçok nörolojik işleve sahip olan
bir nörotransmitterdir ve beyinde yüksek konsantrasyonlarda bulunur. Sinir hücrelerinin
uyarılması ile presinaptik nörondan sinaptik boşluğa glutamat salınır ve postsinaptik
nörondaki glutamat reseptörleri uyarılır. Glial hücrelerde ve nöronlarda da bulunan glutamat
taşıyıcılarının sinaptik boşluktan glutamatı toplaması ile iletim sonlanır. Sinaptik boşluktaki
glutamatın büyük çoğunluğu Glutamat Transporter 1 (GLT-1) tarafından toplanmaktadır.
Glutamat taşınım yolağında, Protein Kinaz C (PKC), Nedd4-2 ubiquitin ligazı fosforile
ederek aktive etmekte, Nedd4-2, GLT-1’i ubiquitine etmekte ve GLT-1 degrade olmaktadır.
Bu proses, GLT-1’in hücre yüzeyi ekspresyonunun, dinamik olarak değişimini ve kontrol
edilmesini sağlamaktadır. Sinaptik boşlukta glutamat birikimi olmadığı durumda, GLT-1
ubiquitinasyona uğrar, yıkılır ve dolayısıyla yüzey ifadesi azalır. Sinaptik boşlukta glutamat
biriktiğinde ise, GLT-1 yüzey ifadesi artar. Bu dinamik prosesdeki bozukluklar, hücreler
arasındaki düşük glutamat seviyesini artırarak, beyin hastalıklarında görülen eksitotoksisiteye
neden olur. Sirtüinler, hücre içindeki çeşitli proteinleri posttranslasyonel modifikasyonlar için
hedef alan protein deasetilaz enzim sınıfıdır. Yalnızca deasetilasyon değil, ADP-ribozilasyonu
gibi posttranslasyonel modifikasyonları da gerçekleştirirler. Sirtuinlerin, nörodejeneratif
hastalıklarda rollerinin bulunduğu bilinmektedir. Sirt4'ün, glioma hücrelerinde, glutamat
metabolizmasını modüle ederek eksitotoksisiteyi engellediği daha önceki çalışmalarda
gösterilmiştir. Ayrıca, Sirt4’ün yokluğunda, GLT-1’e bağlı glutamat alınımının azaldığı
bulunmuştur. Bu çalışmada amacımız, Sirt4’ün, glutamat metabolizmasını ve GLT-1’in

işleyişini nasıl kontrol ettiğini araştırmak ve glia ve glioblastoma hücrelerinde karşılaştırmalı
olarak incelemektir. Bu nedenle, IHA (immortalized human astrocytes-glia) ve U87
(glioblastoma) hücre hatlarında Sirt4 ve GLT-1 protein ifade seviyeleri incelenmiştir. GLT-1
yıkım yolağında görev alan Ubiquitin ve Protein Kinaz C (PKC) proteinlerinin ekspresyon
seviyeleri, western blot tekniği ile incelenmiştir. Ayrıca, glia ve glioblastoma hücrelerinde,
medyumda birikmiş olan glutamatın seviyesi, glutamat assay ile ölçülmüştür. Çalışma
sonucunda, glioblastoma hücrelerinde ubikitinasyonun istatiksel olarak anlamlı olmasa da
azaldığı ve GLT-1’in arttığı görülmüştür. GLT-1 seviyesini kontrol ettiği daha önceki
çalışmalarda gösterilmiş olan Sirt4 ise beklenilen şekilde artmıştır. PKC protein seviyesi ise,
glioblastoma hücrelerinde istatiksel olarak anlamlı olmasa da azalmıştır. Glutamat assayde,
glioblastoma hücrelerinde salınmış glutamat seviyesi, glia hücrelerine göre azalmış olarak
gözlenmiştir. Bunun nedeninin, glia hücrelerine oran ile büyük bir hızla çoğalan U87
hücrelerinin, hiç medyum değiştirilmeden yapılan bu assayde, yüksek oranda ölüm
gösterdikleri için total glutamat salınımının daha az olması olabileceği düşünüldü. Ayrıca,
Sirt4 ve GLT-1, glioblastomada artış gösterdiği için, birikmiş glutamatın toplanarak azalmış
olması, beklenen bir sonuçtur. Bu çalışmada elde edilen sonuçlar, glioblastomada,
eksitotoksisitenin GLT-1 yıkım ya da aktivasyon yolağının modüle edilerek
engellenebileceğini, dolayısyla, yeni ilaç ve tedavi geliştirme alanları oluşabileceğini
göstermektedir.
Glioblastoma multiform is a primary brain tumor derived from glial cells. The tumor growth
occurs via death of neurons surrounding glioma cells dying due to excitotoxicity.
Excitotoxicity is caused by the excessive accumulation of glutamate in the synaptic cleft and
observed in brain diseases such as epilepsy, stroke, and neurodegenerative diseases, as well as
brain tumors such as glioblastoma. Glutamate is a neurotransmitter that has many
neurological functions in the central nervous system and is found in high concentrations in the
brain. After the stimulation of the nerve cells, glutamate is released from the presynaptic
neuron into the synaptic cavity resulting in the induction of the glutamate receptors on the
postsynaptic neuron. Glutamate neurotransmission is terminated in the synaptic cleft after the
clearance of glutamate via glutamate transporters, which are also present in glial cells and
neurons. The vast majority of glutamate in the synaptic cavity is collected by glutamate
Transporter 1 (GLT-1). In the glutamate degradation pathway, Protein Kinase C (PKC)
phosphorylates Nedd4-2 ubiquitin ligase, activated Nedd4-2 ubiquitin ligase ubiquitinates and
GLT-1 is degraded. This process allows for the control of the dynamic cell surface expression
of GLT-1. When there is no glutamate accumulated in the synaptic cleft, GLT-1 undergoes
ubiquitination and breaks down, and hence its surface expression is reduced. When glutamate
accumulates in the synaptic cavity, the surface expression of GLT-1 increases. Dysfunction of
this dynamic process leads to the low level of glutamatein the synaptic cleft causing
excitotoxicity and brain diseases. Sirtuins are a class of protein deacetylase enzymes that
target various proteins within the cell for posttranslational modifications. They perform not
only deacetylation but also posttranslational modifications such as ADP-ribosylation. Sirtuins
are known to have roles in neurodegenerative diseases. Sirt4 has been shown in previous
studies to inhibit excitotoxicity in glioma cells by modulating glutamate metabolism.

Furthermore, in the absence of Sirt4, GLT-1-linked glutamate uptake was found to be
reduced. Our aim in this study is to investigate how Sirt4 controls glutamate metabolism and
the functioning of GLT-1 and also to examine and compare it in glia and glioblastoma cells.
Therefore, Sirt4 and GLT-1 protein expression levels were studied in IHA (immunized human
astrocytes-glia) and U87 (glioblastoma) cell lines. Expression levels of Ubiquitin and Protein
kinase C (PKC) proteins involved in the GLT-1 degradation pathway have been demonstrated
by the western blot technique. Also, in glia and glioblastoma cells, the level of glutamate
accumulated in the medium was measured with the glutamate assay. As a result of the study,
ubiquitination was reduced in glioblastoma cells although not statistically significantly and
GLT-1 was increased. Sirt4, which has been shown in previous studies to control the level of
GLT-1, was found to be increased as expected. The protein level of the PKC protein, on the
other hand, was decreased in glioblastoma cells although not statistically significant.
Glutamate assay showed that the levels of accumulated glutamate in the medium in
glioblastoma cells were reduced compared to glial cells. The reason for this may be that U87
cells, which multiply much more rapidly compared to glia cells, have reduced levels of
glutamate released, as they show a higher rate of death in this assay which was conducted
without changing the medium. Additionally, since Sirt4 and GLT-1 protein levels were
increased in glioblastoma cell line, the accumulated glutamate may have been reduced by the
clearance from the medium by GLT-1. The results obtained in this study show that
excitotoxicity in glioblastoma can be prevented by modulating the GLT-1 degradation or
activation pathway, thus creating new areas of drug and treatment development.
