Bu çalışmada farklı konsantrasyonlarda siklofosfamid (CP) verilen ratlarda sodyum selenitin karaciğer ve böbrek dokularında metallothionin (MT) ekspresyonu üzerine olan etkinliği araştırıldı. Bu amaçla çalışmada toplam 48 adet, ağırlıkları 200±10 gram olan erişkin Wistar Albino ırkı dişi rat kullanıldı. Çalışmada gruplandırma; Grup I (Kontrol grubu; diğer gruplarda patolojik ve immunohistokimyasal değerlendirmeleri karşılaştırmak amacıyla uygulama süresi boyunca hiçbir şey uygulanmadı), Grup II (CP grubu; bu gruptaki deney hayvanlarına 1 gün süre ile her hayvana intraperitoneal olarak 200 mg/kg/gün CP uygulandı), Grup III (Sodyum Selenit grubu; bu gruptaki deney hayvanlarına 1 gün boyunca her hayvana intraperitoneal olarak günlük 1 mg/kg dozda sodyum selenit uygulandı), Grup IV (Sodyum Selenit grubu + CP grubu; bu gruptaki deney hayvanlarına 1 gün süre ile her hayvana günlük 1 mg/kg sodyum selenite ilaveten, 200 mg/kg/gün dozda CP intraperitoneal olarak uygulandı), Grup V (CP grubu; bu gruptaki deney hayvanlarına 35 gün süre ile her hayvana intraperitoneal olarak 5 mg/kg/gün CP uygulandı), Grup VI (Sodyum Selenit grubu; bu gruptaki deney hayvanlarına 35 gün boyunca her hayvana intraperitoneal olarak günlük 1 mg/kg dozda sodyum selenit uygulandı), Grup VII (Sodyum Selenit grubu+CP grubu; bu gruptaki deney hayvanlarına 35 gün süre ile her hayvana günlük 1 mg/kg intraperitoneal sodyum selenite ilaveten, 5 mg/kg/gün dozda CP intraperitoneal olarak uygulandı) ve Grup VIII (Kontrol grubu; bu gruptaki deney hayvanlarına 35 gün süre boyunca patolojik ve immunohistokimyasal değerlendirmeleri karşılaştırmak amacıyla uygulama süresi boyunca hiçbir şey uygulanmadı) olmak üzere 8 farklı grup ve her grupta da 6 hayvan olacak şekilde yapıldı. Çalışmanın sonunda alınan karaciğer ve böbrek örnekleri histopatolojik, ve immunohistokimyasal incelemeler için kullanıldı. Yüksek dozda CP ile birlikte eş zamanlı olarak sodyum selenit verilen gruplarda histopatolojik bulguların şiddeti, yalnızca CP verilen gruba göre anlamlı bir şekilde düşük bulundu. Ayrıca bu gruplardaki dokulara ait MT ile ilgili immunohistokimyasal bulgular ile histopatolojik bulguların şiddeti değerlendirme açısından paralellik gösterdi. Düşük dozda 35 gün süre ile CP ile birlikte eş zamanlı olarak sodyum selenit verilen gruplarda histopatolojik ve immunohistopatolojik bulguların şiddeti, yalnızca CP verilen gruba göre düşük olmasına rağmen istatiksel olarak anlamlı değildi. Çalışmamızda bir gün süre ile yüksek dozda uygulanan CP’nin toksik etkilerine karşı, kuvvetli antioksidan özelliklere sahip sodyum selenit’ in koruyucu alternatif bir kimyasal madde olarak kullanılabileceği görüldü.
The aim of this study was to investigate the protective effects of Sodium Selenite, which have strong antioxidant properties, against the toxic effects of high dose cyclophosphamide on liver and kidney of rats. Fort this purpose, overall 48 adult Wistar albino female rats at the weight of, 200±10 gr were used. These animals were allotted into following groups: Group I (Control group; 1ml/day serum physiologique was administered), Group II (Cyclophosphamid group; each animal was administered daily 200 mg/kg/day cyclophosphamide intraperitoneally for one day), Group III (Sodium Selenite group; each animal was administered daily 1 mg/kg/day Sodium Selenite intraperitoneally for one day), Group IV (Cyclophosphamid+Sodium Selenite group; each animal was administered daily 200 mg/kg/day cyclophosphamide intraperitoneally and 1mg/kg/day Sodium Selenite intraperitoneally for one day), Group V (Cyclophosphamid group; each animal was administered daily 5 mg/kg/day cyclophosphamide intraperitoneally for thirty-five days) and Group VI (Sodium Selenite group; Sodium Selenite group; each animal was administered daily 1 mg/kg/day Sodium Selenite intraperitoneally for thirty-five days), Group VII (Cyclophosphamid+Sodium Selenite group; each animal was administered daily 5 mg/kg/day cyclophosphamide intraperitoneally and 1 mg/kg/day Sodium Selenite intraperitoneally for thirty-five days), Group VIII (Control group; 1ml/day serum physiologique was administered) and each of these eight groups included six rats. After this procedure, followed by euthanasia, tissue samples obtained from liver and kidney, were used for pathological examinations. Tissue samples obtained for pathological examination fixed in 10% buffered formalin solution. Paraffin blocks prepared after routine procedure was cut at the thickness of 4-6 micron and were stained by haematoxyline-eosin (HE) and examined under light microscope. The results of the present study is important in that it investigates the employment of alternative protective method against the toxic effects of cyclophosphamide at high doses. In this context, the fact that Sodium Selenite has no harmful effects, It was tolerated well by the body and It has low cost suggests that the findings obtained might be important for human and animal health.
