Aspergillus flavus HBF34 susu tarafından fermentasyon ortamında üretilen GA
(Glukoamilaz) nisasta afinite kromatografi teknigi ile saflastırıldı. Saflastırılan GA’nın
optimum pH ve sıcaklıgı ile stabiliteleri saptandı. GA’nın farklı substratlar için Km ve
Vmax degerleri ve substrat spesifiteleri bulundu. GA aktivitesi üzerine metal iyonları,
inhibitörler, denatürantlar ve tuz tolerans testi uygulandı. GA’nın ham nisasta
adsorbsiyonu saptandı. _nce tabaka kromatografisi ile amilazın tanımlanması yapıldı.
Ayrıca PAGE, SDS-PAGE ve zimografi ile enzimin kaç adet oldugunu ve molekül
agırlıkları saptandı. Bulunan sonuçlar ilgili literatürlerle karsılastırılarak GA’nın sahip
oldugu birçok üstünlüklerden dolayı endüstriyel uygulamalarda kullanılabilecegi
sonucuna varıldı.
GA starch produced by Aspergillus flavus HBF34 strain in fermentation medium
was purified by affinity chromatography technique. Optimal pH and temperature of
purified GA as well as its stability were determined. Km and Vmax values of GA for
different substrates and its substrate specificity were estimated. Metal ions, inhibitors,
denaturants and salt tolerance test were applied on the activity of GA. Raw starch
adsorption of GA was determined. Identification of the amylase was carried out by thin
layer chromatography. In addition, number of enzymes and their molecular weight were
identified using PAGE, SDS-PAGE and zymography. By comparing with the related
literature, the results gained have suggested that GA could be used in the industrial
applications due to the features of GA.
