Obezite tüm toplumlarda çocuklarla beraber erişkinleri de etkileyen kronik bir hastalıktır. Obezitenin sinir sisteminde, özellikle merkezi sinir sisteminde yaptığı hasar nedeni ile bilişsel fonksiyonları bozduğu bildirilmektedir. Obezite, serebrovasküler patolojiler yanında nörodejeneratif bozukluklara da yol açabilmektedir. İyonotropik glutamat reseptörleri olan NMDA reseptörlerinin öğrenme ve bellek oluşumuna aracılık ettikleri, ayrıca demans ve AH patogeneziyle ilişkilerinin olduğu belirtilmektedir. Bu çalışma yüksek yağ içerikli diyet ile beslenip obez yapılan sıçanlarda öğrenme ve bilişsel fonksiyonların azalıp azalmadığını araştırmak amacıyla düzenlenmiştir. Biz çalışmamızda obezitenin öğrenme, bilişsel fonksiyonlar ve hipokampus üzerine negatif etkisinin olduğu hipotezini araştırmayı amaçladık. Deneysel uygulama için; çalışmamızda sıçanlar herhangi bir ilaç ilavesi yapılmadan %40‟lık yüksek kalorili diyet ile obez yapıldı. Kontrol grubuna ise standart pellet yem verildi. Obezite grubu; erkek (n=15). Kontrol grubu; erkek (n=10) olacak şekilde iki gruba ayrıldı. Lee indeksi hesaplaması kullanılarak obez olduğu belirlenen sıçanlar yer bulma öğrenmesinin değerlendirilmesi için Morris su tankında testlere tabi tutuldu. Kontrol ve obez deney grubu sıçanlarda, platformu bulmak için geçirilen toplam süre değerlerinin istatistiksel olarak grup içi ve gruplar arası karşılaştırması yapılıp obezitenin yer bulma öğrenmesi üzerindeki etkisi değerlendirildi. Morris su labirentinde karşılaştırılan hedef kadranı bulma süreleri açısından gruplar arasında istatistiksel olarak anlamlı bir fark saptanmıştır. Deney bitiminde sıçanlar dekapite edilerek beyin dokularından hipokampüs dokusunda western blot yöntemi ile NMDA reseptör düzeylerine bakıldı. NR2A homojenatlarında kontrol grubu ile obez uygulaması arasında anlamlı bir fark bulunmamıştır. NR2B homojenatlarında ise deney ile kontrol grubu arasında anlamlı fark bulunmaktadır. Elde ettiğimiz bulgulara göre yüksek kalorili diyet ile beslenen sıçanlarda gelişen obezitenin öğrenme ve bilişsel fonsiyonlarda negatif etki yaptığı gözlemlenmiştir.
Obesity is a chronic disease affecting adults and also children in all societies. Obesity is reported to damage cognitive functions due to damage to the nervous system, especially the central nervous system. Obesity can lead to cerebrovascular pathologies as well as neurodegenerative disorders. It is stated that NMDA receptors, which are ionotropic glutamate receptors, mediate learning and memory formation, as well as it is known that they have relationship with dementia and AD pathogenesis. This study was designed to investigate whether learning and cognitive functions are reduced in obese rats fed a high fat diet. We aimed to investigate the hypothesis that obesity is a negative effect on learning, cognitive functions and hippocampus in our study. In our study, rats were fed 40% high calorie diet with before any drug was added for experimental application to get the rats obese. For the control group, standard pellet feed was given. Obesity group; male (n = 15). Control group; male (n = 10). Rats determined to be obese using the Lee index calculation and were subjected to tests in a Morris water tank to assess spatial learning. In the control and obese experimental group rats, the total time spent to find the platform was statistically compared in the groups and inter-groups and the effect of obesity on spatial learning for finding platform was assessed. A statistically significant difference was found between the groups in terms of the time to find the target quadrants compared in the Morris water maze. At the end of the experiment, the rats were decapitated and the NMDA receptor levels were measured by western blot method from the brain tissues to the hippocampus tissue. There was no significant difference between control group and obese application in NR2A homogenates. In NR2B homogenates, there is a significant difference between experiment and control group. According to our findings, it was observed that obesity developed in rats fed with high calorie diet had a negative effect on learning and cognitive functions.
