Kanatlı kan serumlarında mycoplasma gallıseptıcum ve mycoplasma synovıae antikorlarının elısa ile tespiti

Abstract

Bu çalışmada Denizli, İzmir, Manisa ve Aydın İlleri'ndeki broyler civciv ve yumurtacı tavuklarda M. gallisepticum ve M. synoviae infeksiyonlarının serolojik olarak varlığının ve yaygınlığının tespit edilmesi amaçlandı. Çalışma materyalini, aşısız Ross 308 broyler ve Brown Nick yumurtacı tavuklardan, tesadüfi örnekleme yolu ile 60 kümesten alınan toplam 1060 kan örneği (700 broyler ve 360 yumurtacı ) oluşturdu. Serum örneklerinde M. gallisepticum ve M. synoviae'e karşı spesifik antikor varlığı ve antikor titreleri ayrı ayrı olarak ELISA kiti (M. gallisepticum Kit ve M. synoviae test Kitleri, BioChek, Gouda, Hollanda) ile incelendi. Antikor titreleri ELISA kitini üreten Biochek Firması'na ait yazılım programı ile hesaplandı. İzmir, Manisa ve Aydın İlleri'ndeki 700 broyler tavukta M. gallisepticum ve M. synoviae seropozitiflik oranlarının gruplar arası istatistiksel karşılaştırılması için Ki-Kare testi kullanıldı. Her üç ildeki 700 broylerin %26,4'ü M. gallisepticum, %60,2'si M. synoviae antikorları yönünden pozitif olarak tespit edildi. İllere göre M. gallisepticum ve M. synoviae'ye ait seropozitiflik oranları arası fark istatistiksel olarak önemli (P<0,001) bulundu. M. gallisepticum ve M. synoviae yönünden incelenen iller arasında seropozitiflik açısından en yüksek değere sahip ilin Manisa olduğu belirlendi. Çalışma sonuçları alınan serumlara göre değerlendirildiğinde: broyler ve yumurtacılardan alınan toplam 1060 serum örneğinin 419'u (%39,5) M. gallisepticum, 720'si (%67,9) M. synoviae seropozitif olarak belirlendi. Bu sonuçlar, işletmelerde özellikle M. synoviae infeksiyonlarının yaygın olarak görüldüğünü ortaya koydu. İşletmelerde mikoplazma negatifliğinin belirlenmesi amacı ile tarama testlerinin belirli aralıklarla yapılması ve "Mikoplazma Kontrol Programları"nın oluşturulması gerektiği sonucuna varıldı.

The determination of the presence and the prevalence of M. gallisepticum and M. synoviae infections in broiler and layer poultry houses at around Denizli, İzmir, Manisa, and Aydın provinces was aimed in this study. The material was consisted of 1060 blood samples (700 broiler and 360 layer chickens) that had been randomly collected from 60 poultry houses rearing Ross 308 and Brown Nick chickens with no vaccines. The presence of spesific antibody and antibody titers for M. gallisepticum and M. synoviae in sera samples were separately investigated with ELISA (BioCheck, Gouda, Holland). Antibody titers were calculated with a software provided by BioCheck. Chi-Square test was used for seropositivity ratio comparisons between groups. For all broilers in three provinces, seropositivity ratio for M. gallisepticum and M. synoviae was found as 26,4% and 60,2%, respectively. Seropositivity ratios of these provinces were showed statistically significant differences (P<0,001). It was determined that Manisa has the highest seropositity ratio among all provinces. On the other hand, seropositivity ratios for all sera samples (for both broiler and layer poultry houses) were found as 39,5% and 67,9% for M. gallisepticum and M. synoviae, respectively. It concluded that M. synoviae infection was common in these farms. For further investigations, it was recommended that related tests should be used for detection of mycoplasma negativity and Mycoplasma Control Programs should also be formed.
