Ege ve Marmara bölgelerindeki zeytin fidanlık ve ağaçlarında görülen bazı virüs hastalıklarının ELISA testi ile saptanması

Abstract

Bu çalışmada virüssüz zeytin fidanı üretimini gerçekleştirebilmek için üretim materyalinin virüs hastalıkları açısından temiz olup olmadığının Das-Elisa testi ile saptanması amaçlanmıştır. Bu maksatla Balıkesir İlinin Edremit ve Gömeç, Bursa İlinin Orhangazi ve Aydın İlinin Sultanhisar ve Bozdoğan İlçelerinde bulunan zeytin fidanlıklarından 2000 yılı Mayıs, Haziran ve Eylül aylarında alman 220 adet uç sürgün ve genç yaprak örneğine strawberry latent ring spot nepovirus, cherry leaf roll nepovirus, arabis mosaic nepovirus ve cucumber mosaic cucumovirus antiserumlarıyla uygulanan Das-Elisa testlerinde pozitif sonuç alınamamıştır. Bu nedenle yukarıda sözü edilen alanlardaki zeytin fidanlarının üretimi için çeliklerin alındığı anaçlık materyallerden 2001 yılı Mayıs ve Haziran aylarında toplanan 176 adet çiçek örneğine uygulanan Das-Elisa testleri sonucu 64 örneğin (% 36.36) cucumber mosaic cucumovirus, 60 örneğin (% 34.09) cherry leaf roll nepovirus, 69 örneğin (% 39.20) strawberry latent ring spot nepovirus ve 47 örneğinde (% 26.70) arabis mosaic nepovirus ile bulaşık olduktan tespit edilmiştir. The purpose of this study was to determine whether the olive propagation materials are infected with the virus diseases using the Das-Elisa test in order to obtain virus-free olive nursery tree. For this purpose, samples of 220 young leaves were taken from the field nursery in Edremit and Gömeç (Balıkesir), Orhangazi (Bursa), Sultanhisar and Bozdoğan (Aydın) in May, June and September, 2000. The antiserum of strawberry latent ring spot nepovirus, cherry leaf roll nepovirus, arabis mosaic nepovirus and cucumber mosaic cucumovirus applied to 220 samples by DAS-ELISA and the positive result could not be obtained. Because of the above results from the field nursery, 176 flower samples were taken from olive trees in May and June 2001, and Das-Elisa test were applied to the samples. As a result, 64 (36.36 %), 60 (34.09 %), 60 (39.20 %), and 47 (26.70 %) samples were infected with cucumber mosaic cucumovirus, cherry leaf roll nepovirus, strawberry latent ring spot nepovirus, and arabis mosaic nepovirus, respectively.
