Akut faz cevap infeksiyöz, immunolojik, neoplastik, travmatik ya da paraziter
olarak ortaya çıkan nonspesifik bir reaksiyondur. Akut faz reaksiyonun en önemli özelliği,
karaciğerden C-reaktif protein (CRP) , seruloplazmin, fibrinojen, haptoglobin, serum
amiloid A ( SAA) gibi akut faz proteinlerinin üretimidir.
Bu çalışmada ratlarda deneysel olarak oluşturulmuş farklı yangı modellerinde akut
faz proteinleri düzeyleri araştırılmıştır. Akut noninfeksiyöz yangı modeli oluşturmak
amacıyla 0,5 mg/kg dozda turbentin yağı 1. gruptaki ratlara derialtı olarak tek doz
uygulanmıştır. İnfeksiyöz yangı modeli için 106 CFU Staph. auresus yine derialtı yolla 2.
grupta bulunan 21 hayvana, kontrol grubu olarak aynı sayıda hayvana deraltı serum
fizyolojik verilmiştir. Ayrıca çıkış değerleri olarak sağlıklı 7 hayvandan kan örnekleri
hiçbir uygulama yapmadan toplanmıştır. 1., 4. ve 7. günlerde her gruptan rastgele seçilen 7
hayvandan kan örnekleri eter anestezisi altında kalp içi alınmış ve servikal dislokasyon ile
ötenazi uygulanmıştır. Elde edilen tam kan örneklerinden fibrinojen analizi ve tam kan
sayımı; serum örneklerinden CRP, haptoglobin, SAA ELISA yöntemi ile seruloplazmin
analizi, albümin ve total protein analizleri spektrofotometrik olarak yapılmıştır. Ayrıca
serum protein fraksiyonları da elektroforetik olarak gösterilmiştir.
Bu çalışmada haptoglobin, fibrinojen ve seruloplazminin ratlarda akut infeksiyöz ve
noninfeksiyöz yangılarda günler arası değerlendirme yapıldığında uygulama öncesine göre
istatistiki olarak anlamlı düzeyde yükseldiği belirlenmiştir. CRP ve SAA ise uygulama
öncesine göre yükselmiş ancak istatistiki bir önem saptanamamıştır. Ayrıca haptoglobin ve
fibrinojenin infeksiyöz grupta daha yüksek olduğu da gözlenmiştir.
Acute phase reaction is a nonspesific reaction that occurs infectious, immunulogic,
neoplastic, traumatic or parasitic causes. Most important features of acute phase reaction is
the production of some acute phase proteins such as CRP, SAA, haptoglobin,
ceruloplasmin, fibrinogen in liver.
In this study, it was investigated that the levels of acute phase proteins in rats with
experimentally induced different inflammation models. In first groups rats were treated
with 0,5 mg/kg turbentin oil (single dose, ip) for noninfectious inflammation. For
infectious inflammation model, 21 rats that were in second group were treated by 106 CFU
Staph. auresus (ip). 21 rats in control group were given FTS in same way. Additionally,
blood samples were collected to 7 healty animals for 0 days. Days of 1., 4. ve 7., blood
samples were collected intracardiac way in randomly selected 7 rats under ether anesthesia
and applied othenasia by cervical dislocation. Whole blood samples were used for
fibrinogen analyses and blood counting; serum samples were analysed CRP, SAA,
haptoglobin in ELISA reader, ceruloplasmin, total protein and albumin analyses were
measuring by spectrophotometric methods. Serum protein fractions were determined with
electrophoresis.
As a result of this study, haptoglobin, fibrinogen and ceruloplasmin levels were
significantly increased in rats with acute infectious and noninfectious inflammation group
when compare between blood samplind days and in day before induction. CRP and SAA
levels were increased but were not determined any statistically significance. There were
also observed that haptoglobin and fibrinogen concentrations were higher in infectious.
