Abstract: | İri başlı deniz kaplumbağası (Caretta caretta) tropikal ve subtropikal sularda yayılış gösterir. Akdeniz’de en fazla yuvalama alanları Yunanistan, Türkiye ve Libya’da bulunmaktadır. C. caretta’nın Akdeniz’de Türkiye yuvalama kumsallarında kapsamlı olarak populasyon ve koruma genetiği çalışması yapılmamıştır. Bu nedenle, Türkiye’nin Ege ve Akdeniz kıyıları boyunca on sekiz yuvalama kumsalında tespit edilen yuvalardaki ölü yavrulardan doku örnekleri alınarak mitokondrial DNA’nın kontrol bölgesi (n=256) ve altı mikrosatellitlokusu (n=213) izole edilmiş ve C. caretta yuvalama kumsallarının genetik yapısının belirlenmesi amaçlanmıştır. Toplamda yedi tane mtDNAhaplotipi tespit edilmiştir. Yuvalama kumsalları arasında mtDNA bakımından tespit edilen genetik farklılıklar dişilerin yuvalama bölgesine sadakatini ve dişi bireyler arasındaki genetik yapılanmayı gösterirken, mikrosatellit verileri ise çalışılan yuvalama kumsallarında bireyler arasında genetik yapılanma olmadığını göstermektedir. Bu bulgular yuvalama kumsalları arasında erkek geçişli bir gen akışı olduğunu göstermektedir. Sonuç olarak, mtDNA bakımından Türkiye yuvalama kumsalları beş yönetim birimine ayrılmıştır. nDNA bakımından yuvalama kumsalları genetik bir yapılanma göstermemiştir The loggerhead is distributed in tropical and subtropical waters. Greece, Turkey and Libya are the countries with the densest loggerhead nestingin Mediterranean. No detailed studies are available on the population and conservation genetics of the loggerhead turtles nesting on the coasts of Turkey. We, therefore, aimed to determine the genetic structure of the C. caretta populations at the eighteen nesting beaches a long the Aegean and Keywords: Caretta caretta, mitochondrial DNA control region, haplotype, microsatellite, Mediterranean, Mediterranean cost of Turkey by using the control region of the mitochondrial DNA (mtDNA) (n= 256) and six microsatellite loci(n=213). A total of seven mtDNA haplotypes were determined. The mitochondrial genetic differences among nesting colonies supported the nest site fidelity of females while microsatellites did not show genetic structuring which means male-mediated gene flow among nesting colonies. Inconlusion, based on mtDNA results five managementunits were described in Turkey. No genetic structuring was observed among nesting beaches in terms of DNA. |
