Abstract: Amaç: Bu çalışma ile Türkiye'de yayılış gösteren Apiaceae familyasına ait Lisaea türlerinin kloroplast genomunda yer alan trnL-F, trnL-intron ve matK gen bölgeleri incelenerek moleküler sistematik analizleri yapılmıştır. Materyal ve Yöntem: Çalışmada kullanılan bitki örnekleri DNA izolasyon kiti ile izole edilip ardından üç farklı primer (trnL-F, trnL-intron ve matK) ile PZR işlemi uygulanmıştır. Elde edilen PZR ürünleri her bir primer için ayrı ayrı elektroforez cihazında yürütülmüştür. Elektrofofez işlemi sonucunda jel görüntüsü incelendiğinde bantların varlığı tespit edilmiştir. Kullanılan PZR ürünlerinin seansları yapıldıktan sonra ise MEGA 6.0, BioEdit ve Finch TV programları kullanılarak çeşitli parametreler ile filogenetik analizler yapılmıştır. Bulgular :Çalışmadaki filogenetik ağaçlar ML (Maximum Likelihood) yöntemi ile oluşturulmuştur. Üç farklı primer ile çalışılan DNA örneklerine ait her bir primer için parametreler oluşturulmuştur. trnL-F için oluşturulan ağaç 3 gruba, trnL-intron için oluşturulan ağaç 4 gruba ve matK için oluşturulan ağaç 3 gruba ayrılmıştır. Sonuç: Analizler yapılırken oluşturulan karakter temelli algoritma olan maximum likelihood ağaç topolojisi kullanılarak filogenetik ilişkileri incelenmiş, önceki çalışmalardaki veriler ile kıyaslanmıştır. Bu bilgiler doğrultusunda elde etmiş olduğumuz verilerin güvenilir sonuçlar verdiği doğrulanmıştır.
Objektive: In this study, the trnL-F, trnL-intron and matK gene regions in the chloroplast genome of Lisaea species belonging to the Apiaceae family distributed in Türkiye were examined and molecular systematic analyzes were made. Materyal and Methods: The plant samples used in the study were isolated with a DNA isolation kit and then PCR was applied with three different primers (trnL-F, trnL-intron and matK). The obtained PCR products were carried out separately for each primer in an electrophoresis device. The presence of bands was detected when the gel image was examined as a result of the electrophoresis process. After the sessions of the PCR products used, phylogenetic analyzes were made with various parameters using MEGA 6.0, BioEdit and Finch TV programs. Resulun: The phylogenetic trees in the study were created by the ML (Maximum Likelihood) method. Parameters were created for each primer of the DNA samples studied with three different primers. The tree created for trnL-F was divided into 3 groups, the tree created for trnL-intron was divided into 4 groups, and the tree created for matK was divided into 3 groups. Conclusion: Phylogenetic relationships were determined by molecular systematic analyzes of trnL-F, trnL-intron and matK gene regions of Lisaea genera distributed in Turkey. It has been verified that the data we have obtained in line with this information gives reliable results.
