Abstract: Amaç: Bu tezde lakkaz enziminin gümüş kaplı manyetik nanopartiküllere immobilize edilmesi ve biyoremediyasyon potansiyelinin incelenmesi amaçlanmıştır. Gereç ve Yöntem: Gümüş kaplı manyetik nanopartiküller sentezlenmiş ve MUA ile kaplanarak lakkaz enziminin kovalent immobilizasyonu gerçekleştirilmiştir. Karakterizasyonu için FTIR, TEM, EDX ve ESR tekniklerinden yararlanılmıştır. Serbest ve immobilize lakkaz enziminin aktivitesi incelenmiş, optimum pH ve optimum sıcaklık, kinetik sabitler, ısıl ve işlemsel kararlılıkları belirlenmiştir. Ayrıca immobilize lakkazın elektrokimyasal davranışı ile fenolik bileşik ve boyar madde giderimindeki etkinliği değerlendirilmiştir. Bulgular: Serbest ve immobilize enzim için optimum pH 3,0, sıcaklıklar ise sırasıyla 45°C, 55°C olarak tespit edilmiştir. Km ve Vmax değerlerinin immobilizasyon işlemi sonunda azaldığı görülmüştür. Isıl kararlılık incelendiğinde immobilize enzimin serbest enzime göre daha yüksek kararlılık gösterdiği bulunmuştur. İmmobilize enzimin ard arda 10 kez kullanım sonunda başlangıç aktivitesinin %58,13’ünü koruduğu görülmüştür. İmmobilize enzimin elektrokimyasal aktivitesi ile boyar madde ve fenolik bileşiklerin giderimindeki etkinliğinin literatür ile karşılaştırılabilir olduğu bulunmuştur. Sonuç: Bu tezde lakkaz enzimi başarılı bir şekilde manyetik nanopartiküllere immobilize edilmiş ve bazı toksik maddelerin gideriminde etkili olduğu görülmüştür.
Objective: In this thesis, it is aimed to immobilize the laccase enzyme onto silver-coated magnetic nanoparticles and to investigate its bioremediation potential. Material and Methods: Silver-coated magnetic nanoparticles were synthesized, coated with MUA, and covalently immobilized with laccase enzyme. FTIR, TEM, EDX and ESR techniques were used for characterization. The activity of free and immobilized laccase enzyme was investigated, optimum pH, optimum temperature, kinetic constants, thermal and operational stabilities were determined. Additionally electrochemical behavior of immobilized laccase and phenolic compound and dye removal efficiency of the immobilized laccase was evaluated. Results: The optimum pH for free and immobilized enzyme was found to be 3,0 while the optimum temperatures were 45°C and 55°C, respectively. It was observed that the Km and Vmax values decreased at the end of the immobilization process. When thermal stability was investigated, it was found that, immobilized enzyme showed higher stability than the free. It was found that the immobilized enzyme retained %58.13 of its initial activity after 10 consecutive uses. It was found that the effectiveness of the dye and phenolic compound removal of the immobilized enzyme was comparable with literature. Conclusion: In this thesis, the laccase enzyme was successfully immobilized onto magnetic nanoparticles, and it was observed that immobilized laccase was effective for removal of some toxic matters.
