Abstract: Amaç: Mısır, ülkemizin hemen hemen her bölgesinde yetiştiriliciliği yapılan ender bitkilerdendir. Üretiminde, karşılaşılan hastalık ve zararlılara karşı tohum ilaçları ile ilaçlanmaktadır. Kullanılan tohum ilaçları sistemik olmaları sebebiyle bitki tarafından alınarak bitkinin köklerden yapraklara taşınması oradan da gutasyon sıvısına ulaşarak, bu sıvıyı bal arıları (Apis mellifera)’nın su ihtiyacını gidermek için kullanmalarıyla arılarda ani ölümlere ve koloni kayıplarına sebep olmaktadır. Bu tezde gutasyon sıvısında bulunan thiamethoxam ve cyantraniliprole etkili maddelerinin kalıntıları ve bunların arılarda oluşturacağı toksik etkilerin araştırılması amaçlanmıştır. Materyal ve Yöntem: Mısır tohumları thiamethoxam ve cyantaniliprole etken maddeli tohum ilaçlarıyla ayrı ayrı kaplanarak ekimleri gerçekleşmiştir. Mısır bitkisi ekiminden sonra yeni çıkan yapraklarda günlük gutasyon sıvısı takibi yapılmıştır ve gutasyon sıvısı çıkış süresi boyunca toplanmıştır. Toplanan gutasyon sıvısındaki thiamethoxam ve cyantraniliprole etkili maddeleri QuEChERS metoduna göre ekstrakte edildikten sonra, analizleri LC-MS/MS cihazında yapılmış ve belirtilen etkili maddelerin arılara karşı risk durumu hesaplanmış ve LD50 değerleri saptanmıştır. Bulgular: Gutasyon sıvısı çıkış süresi boyunca thiamethoxam konsantrasyonu en yüksek 2,36 ppm en düşük 0,04 ppm değerde, cyantraniliprole konsantrasyonu en yüksek 30,2 ppb, en düşük 0,27 ppb değerlerinde bulunmuştur. İki konsantrasyonun da zamanla hızla azaldığı saptanmıştır. İki etken maddenin arılara risk durumu incelendiğinde 24 saatte thiamethoxam LD50 değeri 1,234 ng/ arı, cyantraniliprole LD50 değeri ise 0,103 μg/ arı olarak tespit edilmiştir. İki etken maddeninde gutasyon sıvısındaki kalıntı konsantrasyonlarına maruz kalan arıların 4 saat, 24 saat ve 48 saatteki ölüm oranlarına etkisi etkili maddelerin içeriğindeki gutasyon kalıntısına paralel gerçekleşmiştir. Sonuç: Denemede kullanılan sistemik tohum ilaçlarının gutasyon sıvısında tespit edildiği ve uçuşa çıkan bal arılarının gutasyon sıvısındaki insektisitlere maruz kaldığı bu nedenle de bal arıları üzerinde önemli risk oluşturduğu tespit edilmiştir. Elde edilen veriler değerlendirildiğinde farklı gruptan sistemik tohum ilaçlarının denenmesi ve arılara toksikolojik denemelerin arazi koşullarında arttırılması gerekmektedir
Objective: Corn is a rare plant that has been grown almost every region of Turkey. During its production, it is being treated with seed coating pesticides against several diseases and pests. Since the treatments are systemic, the pesticide transported from the roots to the shoots, therefore it reaches to the guttation fluid. Honeybees (Apis mellifera) consumes the contaminated guttation fluid to fulfill their water demand, so that this situation causes sudden death and colony losses. In this thesis, it was aimed to investigate the relationship between residues of thiamethoxam and cyantraniliprole in guttation fluid and their toxic effects on bees. Material and Methods: The corn seeds were coated with thiamethoxam and cyantraniliprole, separately, then planted. After corn planting guttation fluid was followed daily, on newly emerged leaves and guttation fluid was collected during emergence period. Thiamethoxam and cyantraniliprole in guttation fluid was extracted via QuEChERS method, then they were analyzed with LC-MS/MS. Risk situation of before mentioned active substances against bees were calculated and LD50 values were determined. Results: The concentration of thiamethoxam during guttation fluid emergence period was found at maximum 2,36 ppm and minimum 0,04 ppm; cyantraniliprole concentration was found at maximum 30,2 ppb while the minimum is 0,27 ppb. Each concentration was decreased rapidly over time. When the risk against bees was calculated, it was found that LD50 values were 1,234 ng/bee and 0,103 μg/bee for thiamethoxam and cyantraniliprole, respectively. When the bees were exposed to the residue concetrations in the guttation fluid of the two active substances for 4 hours, 24 hours and 48 hours; the mortality rates were found parallel to the residues in guttation fluid. Conclusion: The residues of systemic seed coated pesticides were found in guttation fluid, therefore the foraging bees are exposed to insecticides from the guttation fluid fo this reason this situation poses a significant risk to honey bees. When the obtained data is evaluated, it is found necessary to investigate the effects of seed coated pesticides from different groups on bees and toxicological risk assessment trials should be improved under field conditions.
