dc.description.abstractİnterferon Düzenleyici Faktör (IRF) ailesi interferonların savunma sisteminde düzenlenmesini sağlayan transkripsiyon faktörleridir. İnterferon Düzenleyici Faktör 5 (IRF5 bu ailenin bir üyesi olup çeşitli hastalıklarda rolleri ile ilgili çalışmalar son yıllarda artmaktadır. Oktomer Bağlayıcı Transkripsiyon Faktörü 4 (OCT4), embriyonik kök hücre faktörü olarak bilinir. Bu kök hücre faktörü, normal hücrelerin farklılaşmasının yanı sıra kanserin oluşum ve gelişiminde de oldukça önemlidir. Bu tez çalışmasında bir transkripsiyon faktörü olan IRF5 ve bir kök hücre faktörü olan OCT4’nun prostat kanserindeki rolleri ve protein-protein ilişkisi araştırılmıştır. PC3 insan prostat kanseri hücre kültürü yapılarak, pIRF5 ve pOCT4 plazmidleri kullanılarak IRF5 ve OCT4 proteinleri overekspresyon yapılmıştır. Overekspresyon yapılan örneklerde immunopresipitasyon (IP) yapılarak proteinler western blot ile değerlendirilmiştir. IRF5 ve OCT4 overekspresyon yapılan PC3 prostat kanseri hücrelerinde migrasyon düzeyleri scratch yöntemi ile incelenmiştir. Prostat kanser hücrelerinde IRF5 ve OCT4 proteinlerinin birbirleri ile ilişkisi IP deneylerinde protein bantları ile gösterilmiştir. Prostat kanseri hücrelerinde IRF5’in hücre migrasyonunu anlamlı derecede azalttığı, OCT4’ün hücre migrasyonunu anlamlı derecede arttırdığı tespit edilmiştir. Sonuç olarak, IRF5 ve OCT4 proteinleri birbirleri ile etkileşimde bulunan iki transkripsiyon faktörü olup prostat kanser tedavisi için bu ilişki yeni sinyal yolaklarının ve çeşitli ilaç moleküllerinin gelişiminde önemli katkılar sağlayabilir.tr_TR
dc.description.abstractThe Interferon Regulatory Factors (IRF) family are the transcription factor that enables the regulation of interferons in the defense system. IRF5 is a member of this family whic have been increasing studies on their role in various diseases in recently. OCT4 is known as embryonic stem cell factor. This stem cell factor is important in the formation and development of cancer as well as the differentiation of normal cells. In this thesis, the role of IRF5, a transcription factor and OCT4, a stem cell factor, in prostate cancer, and protein-protein interaction were investigated. PC3 human prostate cancer cell culture was performed, using the pIRF5 and pOCT4 plazmids to overexpress the IRF5 and OCT4 proteins. Immunoprecipitation (IP) was performed in overexpressed samples and proteins were evaluated by western blot. Migration levels were examined with overexpression of IRF5 and OCT4 protein in PC3 prostate cancer cells using scratch assay. The relationship of IRF5 and OCT4 proteins in prostate cancer cells is shown by bands in IP experiments. It has been showed that, OCT4 significantly increased and IRF5 significantly decreased cell migration in PC3 prostate cancer cells. As a result, IRF5 and OCT4 proteins are two transcription factors that interact with eachother and this relationship will contribute to the development of new signaling pathways and various drug molecules for prostate cancer treatment.tr_TR
