Abstract: | Denemede farklı yöntemlerle elde edilen sarımsak ekstraktların Hidrojen
Peroksid’in (H2O2) olusturdugu mutagenik etkiye karsı, antimutagenik etkinligi
arastırılmıstır. Bu amaçla 6 farklı sarımsak ekstraktı (taze sarımsak, taze
kaynatılmıs, mikrodalga fırında ısıtılmıs, toz halde, kaynatılarak toz halde ve çift
santrifüjlü sarımsak ekstraktı), pozitif kontrol olarak Hidrojen Peroksit (H2O2) ve
negatif kontrol olarak da bir gece bekletilmis çesme suyu kullanılmıstır. Deney
materyali olarak kullanılan bakla (Vicia faba L) tohumları, mutagen
uygulamasından sonra sarımsaklardan sulu ekstraksiyon yöntemiyle hazırlanan
farklı konsantrasyonlardaki (0,025 ml/ml ve 0,25 ml/ml) farklı sarımsak ekstrakları
ile muamele edilmistir.
Hidrojen Peroksit (H2O2) muamelesi sonucunda hücre bölünme oranında
meydana gelen azalmanın sarımsak ekstraktı uygulamaları ile artıs gösterdigi ve
kromozomal hasar sıklıgının, farklı yöntemlerle elde edilen sarımsak ekstraktları
uygulamasına baglı olarak degistigi ortaya çıkmıstır. Uygulama grupları içerisinde
taze ve toz halde sarımsak ekstraktı uygulamalarının, diger uygulama gruplarına göre
nispeten daha etkili oldugu, ısıtma islemi uygulanan gruplarda ise bu etkinin çok
belirgin olmadıgı görülmüstür. Uygulanan her iki konsantrasyon karsılastırıldıgında
ise bu degisimlerin, konsantrasyona bagımlı olmaktan ziyade uygulama sekli ile
iliskili oldugu bulunmustur. In this study, the anti-mutagenic activity of the garlic extracts which prepared by different methods was investigated against the mutagenic effects of Hydrogen peroxide (H2O2). For this purpose, six different garlic extract were used. Hydrogen peroxide (H2O2) was used as positive control and one night aged tap water was used as negative control in this study. For this purpose, broad bean (Vicia faba L) seeds which were used as experiment material were treated with different aqueous garlic extracts in different concentrations (0.025ml/ml and 0.25ml/ml) after than mutagen treatment. Garlic extract treatments increased the dividing cell rate (mitotic index), which has been decreased after than Hydrogen peroxide (H2O2) treatment. Chromosomal aberration frequency was changed depending on garlic treatments. Fresh garlic extract and garlic powder extract treatments were more effective than the other garlic extracts. It was found that heated garlic extracts showed slight effects on mitotic index and chromosomal aberrations. When the two treatment concentrations of garlic extracts compared, it was found that these changes were more related with the application method than the treatment concentrations. |
