Abstract: | TAVŞAN FEMUR'UNUN MORFOMETRİK VE BİYOMEKANİK
ÖZELLİKLERİ ÜZERİNE OVARİEKTOMİNİN ETKİSİ
Önemli halk sağlığı sorunlarından birisi olan osteoporozun tanı ve tedavisinde ilerleme
büyük oranda uygun hayvan modellerinin kullanılmasıyla sağlanmıştır. Günümüzde
kullanılan birçok hayvan modeli çalışma sonuçları insanlardaki verilerle uyuşmakla birlikte
osteoporozu tek başına taklit edebilen bir hayvan modeli yoktur. Çeşitli ortopedik
çalışmalarda en fazla kullanılan hayvanlardan biri olan tavşanda postmenopozal osteoporoz
ile ilgili çalışmaların çok yetersiz olduğu dikkati çekmektedir. Bu çalışmada altı aylık yaştan
itibaren 10 aylık döneme kadar normal ve ovariektomi uygulanmış tavşan femur'larının
kemik yoğunluğu, biyomekanik ve morfometrik özellikleri açısından faklılıkların ortaya
konulması amaçlanmıştır. Çalışmada altı aylık 24 adet Yeni Zelanda ırkı dişi tavşan
kullanıldı. Çalışma başında hayvanların karaciğer ve böbrek fonksiyonlarına ilişkin kan
analizleri yapıldı. Daha sonra 12 adet hayvanda ovariektomi uygulanırken, 12 adet hayvanda
da yalancı operasyon ile karın boşluğu açılıp tekrar kapatıldı. Operasyonları takiben ikinci ve
dördüncü aylarda DEXA cihazı (Ge Lunar DPX) ile kemik yoğunluğu belirlendikten sonra
her gruptan altı adet hayvanda femurlar çıkartıldı. Çevresindeki dokular tamamen temizlenen
femur'lar serum fizyolojik solusyunu ile ıslanmış gazlı bezlere sarılarak -25oC de saklandı.
Daha sonra kumpas ile dış morfometrik veriler, bilgisayarlı tomografi (GE Hi Speed) ile elde
edilen kesit görüntülerinden AUTOCAD programı ile kesit kalınlıkları, kesit alanları ölçüldü
ve atalet momenti değerleri hesaplandı. Sonra bu kemiklerden DEXA ölçümü ile direktt
kemik yoğunluğu ölçümü tekrar yapıldı. Trabeküler kemiğin biyomekanik özelliklerini ortaya
koymak için condylus femoris'lerden parçalar kesilerek ?indentasyon testi?, kortikal kemiğin
biyomekanik özelliklerini ortaya koymak için corpus femoris'te ?üç nokta eğme testi?
uygulandı. Çalışmanın sonucunda 6 aylık Yeni Zelanda tavşanlarında ovariektomi
uygulandıktan iki ay sonra osteporotik değişikliklerin başladığı ve dört ay sonra ise
osteporozun geliştiği dansitometrik, morfometrik ve biyomekanik yöntemlerle ortaya
konularak, osteoporozla ilgili deneysel çalışma yapacak araştırmacılar için tavşana ilişkin
değerler sunulmuştur. Much of the progress in prevention and treatment of osteoporosis, which is an important public health problem, is supplied by using the suitable animal models. Although there is a good correlation between the results of human and animal modeling studies, no single animal model can be equivalence for the primer osteoporosis in human. Rabbit is the most popular animal for various orthopedic studies, but it is interesting that there has been found limited number of the studies on primer osteoporosis in this animal. In this study, it was aimed to record the some differences between intact and ovariectomized rabbits in respect to bone mineral density, the biomechanic and morphometric properties of rabbit femur. In this study, 24 New Zealand rabbits at six months age were used. The 12 animals were ovariectomized and the other 12 animals were sham group. In two and four months after the operation bone mineral density was evaluated by DEXA (Ge Lunar DPX) under the general anesthesia in the six animals from each group. Then the animals were sacrificed and the bone mineral density was reevaluated by DEXA on the femora. The morphometric measurements of femur were taken with the aid of digital calipers while the sectional measurements were taken with the aid of AutoCAD program by using images of computed tomography. For the biomechanical properties, the indentation test was applied to the cancellous bone samples from condylus femoris while the three-point bending testing was applied to the cortical bone of corpus femoris. In conclussion, the osteoporotic changes were not idendical at two months, but they were noticeable at four months after the operation in the rabbits. The values of this study reflected the bone mineral density, biomechanic and morphometric properties of the femur in ovariektomized and intact female rabbits. It was hoped that this data has supplied a valuable knowledge for the researchers who may use the rabbits as an animal model in diagnosis and treatment methods for the primer osteoporosis. |
