Mastitisli sütlerden izole edilen stafilokokların biyofilm üretme yeteneğinin fenotipik ve genotipik yöntemlerle araştırılması

Abstract

Bu araştırmada 2015 Temmuz ayında, Aydın ili ve çevresinde bulunan işletmelerden ve köylerden laktasyon döneminde olan 100 ineğin her birinden süt örnekleri tekniğine uygun olarak alınıp Adnan Menderes Üniversitesi Veteriner Fakültesi Mikrobiyoloji Anabilim Dalı Rutin Teşhis Laboratuarına soğuk zincirde getirilerek biyofilm oluşturan stafilokoklar fenotipik ve genotipik yönden araştırılmıştır. Bu çalışmada toplanan 100 örneğin, 77 tanesinde bakteriyolojik üreme gözlenmiş bu 77 örneğin Gram pozitif oldugu gözlenmiştir. CRA 'da yapılan değerlendirmeler sonrasında 77 örneğin 13'ünde (%16.8) biyofilm oluşumu negatif bulunurken, 64'ünde (%83.1) biyofilm oluşumu pozitif bulunmuştur. PCR çalışmalarında, biyofilm pozitif olarak değerlendirilen bu 64 izolatın 28'inde (%43.7) icaA ve icaD geni bulunmuştur. Biyofilm negatif olarak değerlendirilen 13 izolatın tamamında ise her iki gende saptanamamıştır.

This research has been done in july 2015, by collecting the samples of milk from the cows in the period of lactation according to the techniques from 100 businesses and villages around aydin and bringing them to the routine identification laboratory of mikrobiology major science department in faculty of veterinary in adnan menderes university with cold chain and the Stafilococcus maintaining the biofilm have been searched according to the fenotype and genotypes. In this research 77 of the 100 samples were seen bacteriological reproduction and they were all gram pozitif. To the results in CRA, the 13 samples of the 77 (%16.8) the biofilm formation was negative however, in the rest (64 of the 77 )(%83.1) it was positive. In the pcr works, in the 28 izolats of the 64 positive biofilms (%43.7), the icaA and icaD gens were found. In 13 izolats of the 13 negative biofilms (%100) the icaA and icaD gens were both not found.
