dc.description.abstractBu çalışmada, uzun süreli metiyonin uygulanan ratlarda homosistein düzeyindeki artışın kalp dokusu ve eritrosit antioksidan sistem üzerine verdiği hasar ve ekzojen bir antioksidan olan vitamin C'nin oluşan bu hasarı ne oranda etkilediğinin araştırılması amaçlandı. Çalışmaya 3 grupta 36 adet rat alındı. Kontrol grubu her gün normal pelet yem ve çeşme suyuyla ad libitum beslendi. Hiperhomosisteinemi grubunda, L-metiyonin ratlara her gün 1 g/kg, Hiperhomosisteinemi+Vitamin C grubu ratlarına da her gün 1 g/kg dozunda L-metiyonin yanında 150 mg/kg/gün vitamin C 6 hafta boyunca gavaj yoluyla verildi. 6 haftalık denemenin sonunda deneme ve kontrol gruplarındaki hayvanlardan kan örnekleri alınarak elde edilen plazma örneklerinde MDA düzeyleri hemen ölçüldü. Plazma örnekleri elde edilirken GSH-Px, SOD ve CAT enzim aktivitelerinin tayini için eritrositler ayrıldı ve analizler gerçekleştirilinceye kadar -20C de saklandı. Servikal dislokasyon ile ötenazi uygulanan ratlardan kalp dokusu örnekleri alındı ve analizlerin yapıldığı güne kadar -80C de muhafaza edildi. Elde edilen doku homojenatında MDA düzeyleri ölçülürken SOD, GSH-Px, CAT antioksidan enzim aktiviteleri analizleri hem doku örneklerinde hem de eritrosit hemolizatında gerçekleştirildi. Ayrıca serum homosistein düzeyleri ticari Rat Homosistein Kiti kullanılarak ELİSA cihazında ölçüldü. Kontrol ve deneme gruplarının serum homosistein düzeylerinin ortalamaları arasında fark (p<0.001) saptanması deneme gruplarında hiperhomosisteineminin oluştuğunu gösterdi. Hiperhomosisteinemi ve Hiperhomosisteinemi+Vit C grupları arasında ise serum homosistein düzeyleri bakımından istatiksel olarak bir anlamlılık saptanmadı. Hiperhomosisteinemi grubuna ait kalp dokusu ve plazma MDA düzeylerinin, Hiperhomosisteinemi+Vitamin C ve kontrol grubundaki plazma MDA düzeylerinden yüksek olduğu tespit edildi (p<0.01). Kalp dokusu SOD aktiviteleri, Hiperhomosisteinemi+Vitamin C grubundaki ratlara göre sadece metiyonin verilen hiperhomosisteinemi grubunda düşük bulundu (p<0.001). Eritrosit hemolizatı SOD aktivitelerinde metiyoninle birlikte vitamin C verilmesi, Hiperhomosisteinemi+Vitamin C grubunda SOD değerlerini Hiperhomosisteinemi grubu değerlerine göre artırdı (p<0.01). Kalp dokusunda GSH-Px aktivitelerinde, kontrole göre deneme gruplarının GSH-Px aktivitelerinde genel olarak bir azalma tespit edildi (p<0.05). Eritrosit hemolizatındaki GSH-Px aktivitelerinde, Kontrole göre Hiperhomosisteinemi grubunda GSH-Px aktivitesinde bir azalış, Hiperhomosisteinemi+VitC grubunda ise bir artış tespit edildi (p<0.001). Kalp dokusu homojenatında ve eritrosit hemolizatındaki katalaz enzim aktivitelerinde kontrole göre Hiperhomosisteinemi grubunda bir azalma, Hiperhomosisteinemi+Vit C grubunda ise bir artış görüldü (p<0.001). Hiperhomosisteinemi grubunda yüksek MDA değerleri, homosisteinin oksidatif strese sebep olduğunu göstermektedir. Metiyoninle birlikte verilen vitamin C dokuda ve plazmada MDA değerlerini düşürdü. Hemolizat ve doku SOD, GSH-Px, CAT aktiviteleri hiperhomosisteinemi grubunda azalırken, Hiperhomosisteinemi+Vit C grubunda arttı. Metiyoninle birlikte verilen vitamin C'nin serum homosistein düzeyleri artışı veya azalışı üzerine herhangi bir etkisinin olmadığı saptandı. Serum homosistein düzeyleri üzerine tek başına yeterli olmadığı kanısına varıldı.tr_TR
dc.description.abstractIn this study, we researched the damage of increase in homocysteine level over heart tissue and erythrocyte antioxidant system on long term methionine applied rats and effect level of vitamin C on this damage, which is antioxidant. In the study 36 rats in 3 groups were used. The Control group were fed by everyday normal pellet feed and tap water ad libitum. In Hyperhomocysteinemia group, L-methionine were given each day by 1g/kg to the rats, to Hyperhomocysteinemia+Vitamin C group the rats were fed besides 1 g/kg L-methionine, 150 mg/kg/day vitamin C were given for 6 weeks by gavage. After 6 weeks study from control and in experimental groups blood samples were taken. In the plasma samples MDA levels immediately measured. While obtaining plasma samples for GSH-Px, SOD and CAT enzymes activities erythrocyte were seperated and kept at -20 oC till to be analyzed. Heart tissue samples were taken from by cervical dislocation euthanasia applied rats and kept -80 oC until analysis hed been done. While measuring the MDA levels in the sample tissues, SOD, GSH-Px, CAT antioxidant enzyme activities were analyzed both in tissue samples and in erythrocyte hemolysates. Additionally serum homocysteine levels were measured with ELİSA apparatus by using commercial rat homocysteine kit. Determining the difference (p<0.001) between mean of serum homocysteine levels in the control and experimental groups, show that occurred hyperhomocysteinemia in experimental groups. In the terms of the serum homocysteine levels statistically non significant among the groups of Hyperhomocysteinemia and Hyperhomocysteinemia+Vit C. The level of heart tissue and plasma MDA in Hyperhomocysteinemia group were greater than Hyperhomocysteinemia+Vit C and Control group (p<0.01). Heart tissue SOD activities were low just in methionine given Hyperhomocysteinemia group compared to Hyperhomocysteinemia+VitC group (p<0,001). By giving methionine with vitamin C in erythrocyte hemolysates SOD activities increased SOD values in Hyperhomocysteinemia+VitC group compared to Hyperhomocysteinemia group (p<0.01). In heart tissue GSH-Px activities, there were general lowering in experimental groups compared to control group (p<0.05). In erythrocyte hemolysates GSH-Px activities, compared to the control group, GSH-Px activity decrease in Hyperhomocysteinemia group, and increase in Hyperhomocysteinemia+VitC group were observed (p<0.001). In heart tissue homogenates and erythrocyte hemolysates the catalase enzyme activities were decreased in Hyperhomocysteinemia group and increased in Hyperhomocysteinemia+VitC group compared to the control group (p<0.001). The high values of MDA in Hyperhomocysteinemia group shows us that, homocysteine causes oxidative stress. Given with methionine, vitamin C lowered MDA values in tissue and plasma. Hemolysates and tissue SOD, GSH-Px, CAT activities decrease in Hyperhomocysteinemia group but increase in Hyperhomocysteinemia+VitC group. Given with methionine, vitamin C has no affect on serum homocysteine levels ıt has not affective on itself.tr_TR
