Abstract: Bu arastırma Aydın yöresinde yetisen bazı endemik bitkilerden (Crocus olivieri ssp. balansae, Scutellaria orientalis ssp. carica, Scrophularia floribunda ve Hypericum adenotrichum) elde edilen ekstraktların sitotoksik aktivitelerinin belirlenmesi amacıyla yapılmıstır. Bu amaçla cins özellikleri göz önüne alınarak 4 farklı endemik bitki seçilmistir. Seçilen bitkilerden farklı yöntemlerle (petrol eteri, etil asetat, diklormetan ve metanol) elde edilen ekstraktların farklı konsantrasyonları (0,5 mg/ml, 1 mg/ml, 4 mg/ml, 20 mg/ml ve 40 mg/ml) bir lösemi hücre hattı olan HL-60 hücrelerine uygulanarak ekstraktların sitotoksik aktiviteleri arastırılmıstır. Ayrıca sitotoksik aktivite sonuçlarına göre etkili olan 3 adet bitkinin HL-60 hücreleri üzerindeki apoptotik ve nekrotik etkileri de Hoechst 33258/Propidium Iodide (HO/PI) boyama yöntemi ile arastırılmıstır. Çalısma sonucunda seçilen bitkilerden özellikle H. adenotrichum bitkisinin hem sitotoksik aktivite hem de apoptotik etki bakımından ön plana çıktıgı gözlenmistir. Sitotoksik aktivite bakımından S. orientalis ssp. carica ve S. floribunda’ da etkili olmustur. Ancak bu iki bitki apoptotik ve nekrotik etki bakımından degerlendirildiginde, S. orientalis ssp. carica’ nın apoptotik etki bakımından, S. floribunda’dan daha etkili oldugu bulunmustur. S. floribunda ise güçlü sitotoksik aktivite göstermesine ragmen, HL-60 hücreleri üzerinde yüksek oranda nekrotik etkiye yol açmıstır. Çalısma kapsamında seçilen endemik bitkiler sitotoksik aktivite bakımından ilk kez bu tez kapsamında arastırılmıstır. Elde ettigimiz veriler kanser tedavisinde kullanılabilecek yeni bitkisel kökenli bilesiklerin arastırılmasına ısık tutması açısından önem tasımaktadır. Daha ileri çalısmalarla bitkilerden elde edilen etkili ekstraktlar içerisindeki etken madde grupları veya bilesikler belirlenerek hücre içerisinde hangi mekanizmalarla ve hangi düzeylerde etkinlik gösterdikleri ortaya konmalıdır.
This study has been carried out to determine the cytototxic activities of the extracts derived from some endemic plants (Crocus olivieri ssp. balansae, Scutellaria orientalis ssp. carica, Scrophularia floribunda ve Hypericum adenotrichum) that are growing in Aydın vicinity. For this purpose 4 different endemic plants have been selected in consideration of genus properties. The cytotoxic activities of different concentrations (0,5 mg/ml, 1 mg/ml, 4 mg/ml, 20 mg/ml and 40 mg/ml) from various extract types (petroleum ether, ethyl acetate, dichloromethane and methanol) of selected plants has been tested against HL-60 leukaemia cell line. In addition the apoptotic and necrotic effects on HL-60 cells of 3 plants which were effective according to the cytotoxic activity results have been investigated with Hoechst 33258/Propidium Iodide (HO/PI) staining. At the end of the study, it was examined that from the selected plants especially H. adenotrichum is coming on surface in both of cytotoxic activity and apoptotic effect. S. orientalis ssp. carica and S. floribunda has been found also cytotoxic. But, when this two plant have utilized in terms of apoptotic and necrotic effect, it was found that S.orientalis ssp. carica was more effective than S. floribunda, related to apoptotic effect. But S. floribunda showed a high ratio of necrotic effect on HL-60 cells in spite of its strong cytotoxic activity. Selected endemic plants in the scope of the study were investigated for the first time in this Thesis in terms of their cytotoxic activity. The results of this study is important in lighting up the investigation for new plant sources that can be used in cancer therapy. Furthermore the act of mechanisms must be shown and the chemical groups or compounds must be determined in the derived effective extracts.
